pH-Sensitive PEG-P(Asp-Hyd-AMK) Nanomicelles Enable Targeted Delivery of Melatonin Metabolite AMK for Attenuation of Oxidative Stress in Alzheimer’s Disease

Alzheimer’s disease (AD) is characterized by chronic neuroinflammation and oxidative stress, where excessive reactive oxygen species (ROS) activate microglia and amplify inflammatory responses. AMK (N1-Acetyl-5-methoxykynuramine), a melatonin metabolite, retains strong antioxidant and neuroprotective properties, but its delivery to the brain is limited by the blood-brain barrier (BBB). In this study, we developed pH sensitive PEG-P(Asp-Hyd-AMK) nanomicelles, where AMK is conjugated via hydrazone bonds. These nanomicelles protect AMK in circulation and release it under acidic conditions typical of AD-affected brain regions. With excellent permeability and targeted delivery, this platform shows potential as a novel antioxidant nanotherapeutic for AD treatment.
