Two-dimensional infrared (2D IR) spectroscopy has made tremendous progress since its pioneering work by Prof. Robin Hochstrasser in 1998. In the introductory part, a brief overview of 2D IR spectroscopy, covering its principle, history, and experimental methods, will be presented. Recent applications of 2D IR to small molecules and large molecular complexes will follow. For acetic acid dissolved in acetonitrile, 2D IR reveals the presence of two isomeric forms of acetic acid: the syn- and anti-isomers. Besides, it uncovers Fermi resonance within each isomer. In another application, 2D IR studies of perovskite films reveal two distinctive absorption peaks in the C=O stretch region, where the two peaks have not reported before. Moreover, 2D IR studies of the aggregation of human islet amyloid polypeptide (h-IAPP) mutants associated with type-II diabetes show that S20G forms relatively shorter oligomers in the early aggregation phase than wild-type hIAPP, where S20G is known to be more cytotoxic than wild-type hIAPP.
