Senescence-associated skin diseases are increasing with aged people. Aged skin exhibits impaired regeneration and own function. To treat such diseases, targeted removal or rejuvenation of senescent cells—the main drivers of these conditions—has emerged as a novel therapeutic strategy. Therefore, a delivery system targeting cell surface vimentin (CSV), a biomarker of senescent human dermal fibroblasts (HDFs), was developed by incorporating CSV-targeting peptide into HDF-derived nanovesicles with known anti-aging effects. The vimentin-targeting cell-derived nanovesicles (Vim-CNVs) were verified by ¹H NMR and DLS/TEM. Senescent fibroblasts treated with Vim-CNVs showed increased proliferation markers, reduced SA-β-gal activity, and enhanced wound healing. Enhanced wound healing was also observed in aged mouse models treated with Vim-CNVs. In conclusion, this study presents a promising approach for targeted drug delivery to treat age-related skin disorders.
