Hyaluronated Lipid Nanoparticles for Enhanced mRNA Vaccine Delivery

Lipid Nano Particle (LNP) has become an important platform for the development of mRNA vaccines. However, the current clinical use of LNPs, which employ poly(ethylene glycol) for mRNA vaccines and may undergo rapid degradation, has been known to cause side effects in a few patients. In this study, we developed a hyaluronate (HA) conjugated LNP for mRNA vaccine delivery. Several cationic lipids were conjugated to HA, and the resulting HA-LNP conjugates were used to form a complex with eGFP-expressing mRNA as a model vaccine. In vitro experiments showed that the HA-LNP/eGFP complex exhibited high expression of eGFP through HA receptor-mediated endocytosis. Additionally, we developed an HA-mRNA conjugate to form a complex with LNP for mRNA vaccines.
