Dual-functional silica particle platform with immobilized enzyme and conjugated polymer for detecting biomarkers

Diagnosing diseases by detecting biomarkers in body fluids provides advantages compared to conventional disease diagnostic systems in terms of time and cost. Fluorescence-based biomarker detection using enzymatic reactions offers selectivity and rapid response. However, this method involves complex procedures and has low reproducibility due to many components such as enzymes and fluorophores. In this study, we developed a silica particle platform (SPP) that incorporates both enzymes and conjugated polymers (CP) to simplify the analysis. The SPP surface was modified with amine and aldehyde functional groups. The enzyme was immobilized via Schiff base chemistry. The CP was processed into nanoparticles and introduced through electrostatic interactions. Upon the enzymatic reaction of the biomarker, the reduced form of nicotinamide adenine dinucleotide (NADH) was generated, increasing blue fluorescence and decreasing the red fluorescence of the CP.
