Transdermal delivery offers a non-invasive alternative to systemic chemotherapy, particularly for anticancer drugs with poor solubility and limited patient compliance. To address these challenges, we developed ethosomes incorporating paclitaxel and quercetin, two lipophilic agents known for their synergistic anticancer effects. Ethosomes, composed of ethanol-enriched phospholipid bilayers, exhibit excellent deformability and enhanced skin permeability, making them suitable carriers for hydrophobic drugs.

To control the physicochemical properties of the ethosomes, we employed a microfluidic mixer for precise and reproducible synthesis. Transdermal drug permeation was quantified using a Franz diffusion cell to evaluate delivery performance based on vesicle size and composition, followed by in vitro studies to assess therapeutic efficacy. This platform may provide a promising strategy for localized melanoma therapy with improved therapeutic outcomes and reduced systemic toxicity.
