Evaluation of Local Anaesthetic Release from Biodegradable GelMA Beads for Embolization

Gelatin methacryloyl (GelMA) hydrogels have gained significant attention in biomedical applications due to their biodegradability, cytocompatibility, and customizable network structure. In pain management, local anaesthetics such as Ropivacaine (ROP) and Lidocaine (LID) are frequently administered; however, their rapid clearance limits their therapeutic window. A promising strategy to address this involves harnessing pH-responsive electrostatic interactions between drug molecules and hydrogel functional groups. Specifically, the protonated amine group (-NH₃⁺) of anaesthetics exhibits enhanced affinity toward sulfonate groups (-SO₃⁻) under acidic conditions, promoting drug entrapment and sustained release. We investigated and compared the encapsulation efficiency and release behaviour of ROP and LID across different pH environments.
