Polymers can store information with higher storage density and longevity than conventional media. The sequencing of polymers has relied on mass spectrometry, which imposes complexity in sequencing when the multitudes of polymer chains utilized to store information. Here, we demonstrate a decoding and random access of information stored in sequence-defined copolymers, poly(phenyllactic-co-lactic acid) (PcL), composed of up to 256 α-hydroxy acids as monomers having molecular weights far exceeding the detection limit of tandem mass spectrometry. It is based on the fragmentation of PcL into oligomers using scissile monomer implemented at random positions in the PcL chain using fragmentation codes. Mandelic acid was used as scissile monomer. Under a hydrogenolysis condition, PcL is fragmented into oliomers as directed by the number and location of the fragmentation code. Also, a random access is enabled by decoding the fragment containing the target bits without sequencing an entire chain.
