Development of cell/microparticle spheroid system to optimize TGF-b1 concentration in chondrogenic differentiation

Osteoarthritis (OA) is a degenerative disease that causes a significant deterioration in patients’ quality of life. In recent years, cell therapy has been raised as a promising regenerative therapy for OA and cell spheroid has been frequently adopted. However, conventional cell spheroids have limited supply of growth factor (GF) into the central region for chondrogenic differentiation. Therefore, the appropriate GF concentrations to induce chondrogenesis are controversial. In this study, we developed cell spheroids with microparticles with leaf-stacked structure (LSS particles). LSS particles are evenly distributed among cells and provide space for supply of GF into central region of cell spheroid, and thus effectively induce chondrogenic differentiation even at significantly lower GF concentration than the conventionally used. Based on the findings, we suggest that the cell/LSS spheroid can be a platform system which allow effective penetration of bioactive molecules into cell spheroids.
