Bioinspired transdermal delivery patches with multiscale adhesive architectures

Drug delivery through the skin layer is a clinically valuable but challenging task primarily due to the multilayered stratum corneum exhibiting barrier function. This work introduces the design, operation mechanism, and performance of noninvasive cephalopod-limb-like patches with preload-dependent dual-layered suction chamber cluster (d-SCC). Combining the embossed structure with the dome in chamber, the tentacle-like device enables deformation control of stratum corneum of the engaged skin. This innovative design allows for efficient transport of diverse drugs through the complex stratum corneum, without skin discomfort. Compared the data with and without d-SCC, application of its negative pressure to the porcine, human cadaver, and artificial skin significantly improved the penetration depth of liquid-formulated drug. Furthermore, the potential feasibility and efficacy of d-SCC were demonstrated in atopic dermatitis-induced model with thickened stratum corneum, inspiring potential advancements in the medical and skincare fields.
