Hydrogel for Inhibiting Venous Intimal Hyperplasia In Arteriovenous Fistula

Arteriovenous fistula(AVF) is a surgical procedure that connects an artery to a vein for hemodialysis, allowing increase the amount of blood flowing. AVF is one of the favored vascular access, but 60% of AVFs fail to successfully mature due to early intimal hyperplasia development. To overcome vessel occlusion due to intimal hyperplasia, blood vessels must be dilated to prevent a decrease in blood flow rate. Here, we develop injectable hydrogel that effectively deliver vasodilating drug. Hydrogel consists of oxidized guar gum(OGG), gelatin and microbial transglutaminase(mTG) which is FDA approved and biocompatible for intravascular application. The oxidation rate of OGG and the mixing ratio of OGG and gelatin allowed the degree of Schiff base bonding to be controlled, resulting in injectable properties. Sildenafil citrate was used as a drug to promote vascular regeneration, and confirmed angiogenesis and sustained release of drug. Finally, good biocompatibility was verified.
