Developing nanoparticles with analgesic properties for the treatment of Chemotherapy Induced Peripheral Neuropathy (CIPN)

Chemotherapy-induced peripheral neuropathy (CIPN) is a common side effect of chemotherapy resulting in various neurological problems. Aconite root (AR) has been traditionally used for its analgesic properties due to the presence of the active ingredient aconine, which blocks the Na+ channels in the peripheral nerves, reducing pain. However, AR also contains the toxic substance aconitine, which can be reduced through various techniques. One method for reducing aconitine is via boiling, and the optimal time found to be 48 hours. To deliver aconine efficiently, HA-PLGA nanoparticles encapsulated AR were prepared. These nanoparticles demonstrated efficient drug release and establishing effective transdermal drug delivery of aconine. PC12 cells treated with paclitaxel were used to establish a CIPN-like environment, and the neurites derived from HA-PLGA/AR NPs demonstrated an increase in growth compared to the control group, validating the localized analgesic effect of the nanoparticles on CIPN.
