pH- Sensitive Nanoparticle-Incorporated Contact Lens for On-Demand Drug Delivery

We introduce a contact lens with pH sensitivity for on-demand drug release only in wearing situation, with negligible premature drug leakage during packaging and transit, which is accomplished through the incorporation of drug-loaded mesoporous silica nanoparticles (MSNs) coated with a pH-sensitive polymer into the contact lens. The compositionally optimized, pH-sensitive polymer has a lower critical solution temperature (LCST) above 45°C at pH 7.4, whereas its LCST decreases below 35°C in acidic state (pH ~ 6.5). Consequently, the MSN-incorporated contact lens can allow the sustained release of the loaded drugs only at acidic condition and 35°C, which corresponds to the wearing situation, through the MSN pores opened by the shrinkage of the polymer chains, whereas low temperature condition or neutral pH, corresponding to packaging and transit, enables a negligible level of drug leakage.
