Preparation of uniform gene-loaded lipid nanoparticles using membrane extrusion process

With the advent of Covid-19, gene-loaded lipid nanoparticles have been actively investigated due to their advantages, fast development speed and less immune reactions. Lipid nanoparticle (LNP) is a lipid-based carrier that delivers gene into the cell safely and its properties such as size, composition, and morphology are known to determine delivery efficiency. To control these properties, various fabrication methods are used. One of them, the bulk mixing method, is advantageous for large-scale production, but it is difficult to form uniform particles. Here, we proposed a membrane extrusion process to get homogeneous LNPs. Additional extrusion process is conducted after preparing pre-mixed LNPs by simply mixing ethanol containing lipids and water containing gene. Through the membrane extrusion process, we could obtain uniform, uni-lamellar LNPs with a size of 100nm or less.
