The study developed a thermal responsive hybrid nanoparticle (TRH) named F-TRH, which combined photothermal therapy and immunotherapy against cancer and its metastasis. F-TRH, decorated with FimH and containing a near-infrared absorbent, induced cancer cell apoptosis in vitro and showed therapeutic effects against tumors in mice when coupled with NIR laser irradiation. The released FimH and cancer cell apoptotic bodies promoted dendritic cell activation, leading to cancer antigen-specific T cell immunity. As a result, cured mice from 1st challenged tumor by F-TRH prevented the 2nd challenge of CT26 and 4T1 cell growth in the lung, which was mediated by antigen specific T cell immunity. Thus, these data demonstrate the potential use of F-TRH for photothermal-immuno therapy against cancer and its recurrence and metastasis.
