Enhancing Cellular siRNA Delivery via Dendritic Assembly of Gold Nanoparticles

RNA interference (RNAi) has received growing attention for possibility of treat hard to cure disease and gene-related disease in gene therapy due to gene silencing effect. Herein,Successful cellular delivery of synthetic siRNA depends mainly on the capability of a carrier to form a stable complex with siRNA, which can provide efficient protection of the siRNA from enzyme-mediated degradation and improved cellular uptake. In this study, dendritic gold nanoparticles (Au@MC) were applicated for siRNA delivery. The siRNA could be physically localized in the extra space between braches on Au@MC, which leading to efficient cellular uptake and improved gene silencing effect.Among various formulations of dendritic gold nanoparticles, Au@MC-2/siRNA complex exhibited efficient gene suppression in the absence and presence of serum with low cytotoxicity. It is anticipated that particular grooves having Au@MC have a chance at siRNA protection against enzymatic digestion, leading to serum stability and improved gene silencing effect.
