Acid-responsive polymeric micelles with superior stability as immunostimulating anticancer nanomedicine

Coupling of reactive oxygen species (ROS) elevation with glutathione (GSH) depletion amplifies oxidative stress to kill cancer cells selectively and also elicit immunogenic cell death (ICD). Despite these advantages, translation of oxidative stress amplifying compounds has been restricted as these compounds generally require additional drug carriers. Here, we formulated T-POS micelles, composed of oxidative stress amplifying hybrid prodrug (OSamp), amphiphilic poly(ethylene glycol)-block-poly(cyclohexyloxy ethyl glycidyl ether)s (PCHGE) and lipopeptide (DSPE-PEG-RGD). T-POS micelles potently exerted antitumor activity and also induced strong ICD featured by the expression of DAMPs (damage-associated molecular patterns). In CT26-bearing mice, T-POS micelles enhanced immune responses by increasing the population of cytotoxic CD4+ and CD8+ T cells. T-POS micelles pave a promising way for inhibiting tumor growth and amplifying oxidative stress as immunostimulating anticancer nanomedicine.
