Enhanced Cancer Vaccination by Dissolving Microneedles

Microneedle (MN) has considered as a minimally invasive skin drug delivery system that can overcome skin problems. Among them, dissolving microneedles (DMN) have attracted due to their simple fabrication and convenient application. DMN dissolves rapidly upon contact with intradermal fluids, enabling efficient drug delivery and minimizing the risk of inflammation. However, since DMN are formulated from hydrophilic polymers and dissolve rapidly in intradermal fluids after administration, they are often difficult to application with water-insoluble vaccine components. We developed amphiphilic polymer-based DMN and used them as carriers for transdermal delivery of hydrophobic adjuvant (R848) and hydrophilic cancer model antigen (OVA). Application of MN containing R848 and OVA to skin cancer mice of EG7-OVA induced significant levels of antigen-specific humoral and cellular immunity, resulting in significant anti-cancer activity.
