Bioactive HA filler loaded with PLLA-based microspheres that regulates collagen production and angiogenesis

Microsphere-containing HA fillers further improve wrinkles and extend retention among the widely used HA fillers. However, the acidic by-products produced during the decomposition of microspheres composed of biodegradable polymer cause an inflammatory response. In this study, we fabricated poly(L-lactic acid) (PLLA) microspheres contained with magnesium hydroxide (MH) and zinc oxide (ZO) and then prepared a functionalized HA filler including the microspheres and drug. MH has a pH neutralization effect on acidic by-products from biodegradable polymers. NO, which induces wound healing and angiogenesis, is generated by reacting with ZO in the body. A drug has been used to stimulate collagen production and wound repair. The bioactive HA filler reduced the expression of inflammatory genes and upregulated collagen production and angiogenesis. Excellent volume retention was shown in the injected filler subcutaneously in the mice. It is anticipated to become a new technology for facial rejuvenation.
