Inspired by melanogenesis found in nature, we developed tyrosinase-mediated one-pot zwitterionic coatings for anti-biofouling applications. In this study, amine-terminated sulfobetaine (SBAm) and carboxybetaine (CBAm) were synthesized and grafted onto surfaces via Michael addition and Schiff base reaction with enzymatically oxidized tyrosine whose catecholamine group serves as a coating precursor. The surface coatings were characterized by spectroscopic ellipsometry, water contact angle goniometry, and X-ray photoelectron spectroscopy, demonstrating that the coatings can be formed in a substrate-independent manner. Due to the zwitterionic moieties encapsulated in eumelanin, SBAm/CBAm-eumelanin films effectively inhibited nonspecific protein adsorption and bacteria adhesion. We believe that this simple and versatile coating method can be used on demand and has excellent potential for biomedical applications where anti-biofouling is required.
