Effect of aligned magnetic fiber fragments on differentiation of C2C12 myoblast.

C2C12 myoblasts proliferate and differentiate to the precursors of contractile skeletal muscle cells through the process of myogenesis. The degree of differentiation and formation of myotube varies in different environments. So, we confirmed the effect of alignment of myoblast on myotube formation in myogenesis. To make cell encapsulated 3D model, we use gelatin methacrylate (GelMA) and hydrolyzed magnetic fiber fragments. Mechanical property of GelMA and fragments-laden GelMA hydrogels were evaluated using rheometer. Cell proliferation was confirmed by LIVE/DEAD assay. To check the structure of cells, DAPI/F-actin staining was performed. Formation of myotube and other factor was confirmed using immunostaining. This work will find proper environment of culturing C2C12 myoblasts. And it is expected to this matrix can differentiates C2C12 myoblast more efficiently.
