PLGA Microspheres with Magnesium Hydroxide and Vitamin D for Osteoinductive Calcium Sulfate Hemihydrate-based Bone Paste

With the development of minimally invasive approaches, injectable bone paste has attracted interest as a synthetic substitute. Among them, calcium sulfate hemihydrate-based bone paste is biodegradable and has analogous composition with human bone. However, its fast resorption and degradation rate are often criticized as it can be absorbed into the body before new bone formation occurs. To remedy their shortcomings, we combined β-tricalcium phosphate (β-TCP) with α-CSH for slow resorption and osteoconductive capacity. Magnesium hydroxide (MH), an alkaline ceramic, has attracted attention as an additive to improve biocompatibility. Vitamin D is a hydrophobic secosteroid hormone that generally regulates calcium and phosphate metabolism. In this study, we designed a MH/VitD/PLGA microsphere-incorporated α-CSH/β-TCP injectable paste for effective bone regeneration. It induced osteogenic differentiation and regulated inflammation responses which could be generated during the mass bone surgery.
