Designing Bioactive Hydrogels Releasing Magnesium-ions for Facilitated Macrophage Polarization and Neovascularization

An emerging paradigm in designing polymeric hydrogels for tissue regeneration is creating bioactive matrices that stimulate surrounding tissues via various biological and physicochemical stimuli implanted in vivo. Growing evidence demonstrates that magnesium ions (Mg2+) are critical in wound healing and tissue regeneration, such as immunomodulation and neovascularization. Herein, we design Mg2+-releasing hydrogels (Mg-Gel) via magnesium peroxide-mediated crosslinking reaction. Our Mg-Gels are formed via disulfide bonds and thiol-ene reactions between polymer backbones. Our hydrogels have controllable phase transition time and elastic modulus depending on their composition. In addition, our Mg-Gels show excellent cyto- and tissue compatibility. Furthermore, bioactive matrices accelerate skin wound healing by promoting macrophage polarization and neovascularization. In sum, our Mg-Gels have the potential as advanced polymeric hydrogels for tissue regenerative medicine.
