Synthesis and Characterization of Self-Assembled Lipitoid Hydrogel

Peptoids, as sequence-defined polymers, show great promise as a novel hydrogel material candidate due to its exceptional phisical/chemical stability and enzyme resistance, in comparison to peptides. However, their high structural flexibility poses challenges in forming self-assembled nanostructures, leading to limitations in materials applications due to relatively lower mechanical properties. Herein, we developed 'lipitoid,' a peptoid (N-susbsituted glycine oligomer) with alkyl chains at N-terminus for easy self-assembly, inspired by the lipopeptides. Lipitoids demonstrated a significantly faster self-assembly into nanostructures in aqueous solutions compared to conventional peptoids, forming a network that resulted in the formation of a hydrogel. Remarkably, the lipitoid hydrogel exhibits an exceptionally rapid gelation rate upon the introduction of salt. These characteristics make lipitoid hydrogel a promising candidate for various biomedical applications, including use as a material for spraying or printing purposes.
