Surface-enhanced Raman Optical Activity Tuning for Conformation Information of Biomolecules Using Chiral Gold Nanoparticles

Raman optical activity (ROA) has been becoming an effective chiroptical probe for the analysis of the stereochemistry of biomolecules. Despite the intrinsic chirality of biomolecules, a convincing ROA spectrum demands a long collection period, high-powered laser, and short wavelength laser excitation, which causes severe structural damage to biomolecules. Here, we propose chiral gold nanoparticles (AuNPs) that are highly distorted and size-tunable for improving the conventional ROA measurement. Through multiple seed-mediated growths utilizing cysteine, the optical activity of chiral AuNPs is enhanced under the near-infrared region. This work can provide a practical approach for obtaining the conformational data of biomolecules from ROA measurement, which may be applicable for biosensors, disease diagnosis, and other biological devices.
