Isolation of extracellular vesicles using nano-porous membranes with uniform nanopores.

Mesenchymal stem cell-derived extracellular vesicles (EVs) have shown the potential to facilitate wound healing, modulate immune responses, and inhibit inflammatory diseases. However, current methods for isolating EVs from cell culture media pose a challenge to achieving high purity and separation yield for therapeutic applications. Herein, we report a two-step tangential flow filtration (TFF) system with uniform nanoporous track-etched membranes to isolate EVs. After TFF process employing two different nanopore membranes, highly purified EVs were selectively isolated from proteins and micron-sized vesicles through size exclusion mechanism. This purification was confirmed through nanoparticle tracking analysis and measurement of exosome-specific markers. Notably, the structural integrity of the EVs remained intact following the TFF process. These findings hold great promise for advancing the therapeutic applications of EVs in regenerative medicine and immunomodulation therapy.
