A Glycol Chitosan Prodrug Blend Thermogel: A Novel Injectable Formulation for Enhanced Inner Ear Drug Delivery

The current inner ear drug administration methods have unmet essential clinical efficacy and safety standards. This study introduces a novel self-degradable prodrug blend thermogel system for safe and effective inner ear drug delivery, promising significant clinical advancements. The new formulation combines hexanoyl glycol chitosan (HGC) as a thermogel component and dexamethasone-conjugated glycol chitosan (GC-DEX) as a prodrug component, designed with significant thermogelation, injectability, and self-degradation properties, ensuring controlled drug release. The evaluation shows improved drug absorption and sustained inner ear drug concentrations, accelerating hearing loss recovery. Furthermore, the drug release induces thermogel dissociation, enabling self-removal from the middle ear cavity. Consequently, the prodrug blend thermogels are promising candidates as an effective injectable formulation for inner ear treatment, fostering more efficient management of inner ear diseases.
