CHA (catalytic hairpin assembly)-based naked-eye assay for target gene detection

Infectious diseases such as COVID-19 are of major concern worldwide due to their tremendous transmissibility in a short period of time. Accurate, simple, and cost-effective genetic assays for target gene detection are required to effectively prevent the spread of infectious diseases. In this study, we developed a naked-eye assay based on catalytic hairpin assembly (CHA) reaction for simple and sensitive detection of target genes. CHA reaction forms a DNA product through self-assembly between complementary base sequences by specifically reacting to the target gene without enzymatic reaction at room temperature. A small amount of the target gene forms a large amount of DNA product through a cyclic reaction. The resultant DNA product is combined with the naked-eye assay, enabling simple and sensitive naked-eye detection of the target gene. The developed CHA-based naked-eye assay can be used as a platform technology that can quickly and accurately detect various target genes.
