A Reversibly Photo-controllable Hydrogel Based on Hyaluronateand Green Fluorescent Protein for Drug Release

Hyaluronic acid (HA), a natural polysaccharide, was found in the extracellular matrix of articular cartilage and synovial fluid. Several methods for chemical modification of HA were described. Among those biomaterials, hydrogel made of HA is of great interest because of its distinctive features, such as biocompatibility and controllable physical properties. Here, we introduce a novel photo-responsive hydrogel based on maleimide-modified hyaluronic acid (HA-Mal), conjugated by reversibly photo-dissociable dimeric green fluorescence protein, pdDronpa. This hydrogel can undergo reversible transition between gel and sol states upon cyan and violet light illumination, as a result of the trans-cis isomerization of chromophore exists in pdDronpa that weakens and recreates the dimeric interface of pdDronpa. Base on this feature, a new hydrogel platform can be applied to study drug delivery and controlled release of molecules.
