Dexamethasone loaded and biodegradable core-sheath biphasic monofilaments: Drug release and degradation studies

Although biodegradable polymeric filaments have been widely used for biomedical applications, these still have some problems such as inflammation and mechanical properties. In our previous study, poly(L-lactide)-polydioxanone (PLLA-PDO) core-sheath monofilaments have been developed and characterized. In this study, mechanical properties, biodegradability, and the drug release of series of PLLA-PDO core-sheath monofilaments containing dexamethasone (Dex), an anti-inflammatory drug, were investigated. Biphasic structures showed the advancement of mechanical properties in the degradation process. Biodegradation could be controlled by biphasic structure and drawing process. PLLA-PDO monofilaments released Dex in sustained or controlled manners through the drug location in the biphasic structure and drawing rates. In conclusion, our drug-eluting sheath-core monofilament stents can be a promising model for advanced nonvascular stents, the targeted application of this study.
