PAMAM dendrimer with enzyme responsive sequence and cystamine core for gene delivery

In this study, development of gene delivery systems based cystamine core PAMAM dendrimer containing the enzyme sensitive sequence ‘GFLG’ was evaluated to efficient transfection efficiency. Cystamine core of PAMAM generation 3 is responsive to intracellular glutathione, it is the reduction of a disulfide bond. And the surface of cPAMAM G3 was conjugated with ‘GFLG’ peptide sequence, ‘GFLG’ sequence is enzyme responsive sequence by cathepsin B and could be cancer-specific targeting due to cathepsin B overexpressed in cancer. R-H-GFLG-cG3 PAMAM showed for high transfection efficiency and low cytotoxicity in HepG2 and SW480 cells.
