Efficient synthesis of sequence-defined polymers by continuous flow system

Synthesis of a high molecular weight polymer with absolutely defined molecular weights and sequences has immense potential for applications in a wide range of fields. Precise polymer synthesis via cross-convergent method involving numerous unit reactions is time consuming process. Here, we report semiautomated synthesis of sequence-defined polymers through adequate combination of flow chemistry and cross-convergent method. This system enables efficient synthesis by integrating the three reactions, including two deprotection and one coupling reactions, and an in-line purification. Our proposed method allows the production of all kinds of tetramers composed of lactic acid and glycolic acid in a day. The synthesis of a sequence-defined polymer with 64 repeating units is demonstrated. The stored sequence information can be fully retrieved by mass spectrometry.
