Abstract
The use of small interfering RNA (siRNA) in tumor therapy provides a powerful approach for selectively silencing target genes via RNA interference (RNAi). To enhance systemic delivery of siRNA, we developed an advanced carrier system based on low molecular weight polyethylenimine (PEI, 1.8 kDa) conjugated with deoxycholic acid (DA), referred to as DA-PEI. Building upon this foundation, we further optimized the system by integrating 4-fluorothiophenol (4-FTP) into the DA-PEI framework (4-FTP-DA-PEI). This modification leveraged π-π interactions between aromatic rings and coprostane structures to improve stability. Our results demonstrated that 4-FTP-DA-PEI exhibited enhanced serum stability, increased cellular uptake, and higher transfection efficiency. In a mouse tumor model, systemic delivery of 4-FTP-DA-PEI/thiol-siRNA complexes effectively inhibited tumor growth, underscoring the potential of this delivery system as a promising therapeutic platform.
