Abstract
This study designs a glucose-responsive microneedle system by sequentially optimizing PtNP catalytic activity, microneedle fabrication, and release behavior. First, PVP/citrate-stabilized PtNPs will be prepared via ascorbic acid reduction and characterized using TEM, DLS, zeta potential measurements, and UV-vis spectroscopy. Their catalytic function will be evaluated using an H2O2 solution to measure H2O2 decomposition and dissolved O2 generation across different PtNP concentrations and pH values. After selecting the lowest effective PtNP concentration, the microneedles containing GOD and PtNP were prepared. The prepared microneedles will be characterized by optical microscopy, SEM-EDS mapping, CLSM, mechanical strength, in vitro skin permeation, swelling, dissolution, drug loading, and glucose-responsive release.
