Abstract
As the demand for cryogenic storage and transportation of biopharmaceuticals and biological materials continues to increase, cryopreservation has become increasingly important. Cryoprotective agents (CPAs), such as dimethyl sulfoxide (DMSO), are commonly added to cell culture media to reduce cellular damage during freeze-thaw cycles. However, conventional CPAs may cause cytotoxicity and incomplete recovery of cellular function after thawing. In this study, a peptide which previously designed according to the site-distance principle to achieve ice lattice matching, was conjugated to gold nanocubes. Using the resulting peptide-conjugated gold nanocubes, we developed a hybrid colloidal solution for cell cryopreservation. The hybrid gold colloidal solution exhibited ice recrystallization inhibition activity and altered the freezing and melting behavior of cell suspensions. Cell-based evaluations further demonstrated its cryoprotective efficacy, as evidenced by improved post-thaw cell viability and metabolic activity.
