Abstract
Adeno-associated virus (AAV) has emerged as one of the most promising vectors for in vivo gene therapy because of its excellent safety profile, broad tissue tropism, and long-term transgene expression. Consequently, AAV-based therapeutics have achieved remarkable clinical success across a wide range of genetic diseases. Despite these advances, the limited packaging capacity of AAV remains a major obstacle to the delivery of large or functionally complex genetic payloads, often requiring multiple vectors to achieve therapeutic outcomes. Here, we present a programmable strategy for assembling different AAV vectors using our previously developed ultra-site-specific bioconjugation chemistry. This modular approach enables the controlled association of AAV particles carrying different genetic cargos, providing a potential route toward coordinated multivector gene delivery and expanded cargo capacity.
