Abstract

Oral cancer vaccines provide a non-invasive approach to enhance patient compliance. We developed glycocholic acid (GCA) and mannose-coated liposomal nanoparticles (GMN) encapsulating ovalbumin and poly(I:C) for dendritic cell (DC) targeting via oral delivery. Mannose-free GN showed 2-fold higher apical sodium-dependent bile acid transporter-mediated uptake than GCA-free CN. Oral GMN promoted DC maturation, increased CD86, CD40, and CD80 expression, and induced a Th1-skewed response with a 2-fold elevation in the IgG2c/IgG1 ratio. In vivo, GMN completely prevented B16F10 melanoma and reduced CT26 colon cancer incidence by 70%. In the pre-administration group, melanoma did not develop up to day 33. These results indicate that GMN integrates oral bioavailability, lymph node targeting, and immune activation, demonstrating strong tumor-preventive efficacy as a promising oral cancer vaccine platform.
