Abstract

Cryopreservation is a key technology for the long-term storage of biological samples. However, cell viability is affected by ice formation and recrystallization, osmotic stress, and the toxicity of cryoprotectants. Most studies in cryopreservation have concentrated on improving the cryopreservation process, particularly through the development and optimization of cryoprotective agents. Thawing process has received relatively limited attention, even though ice recrystallization during thawing is a critical factor. Therefore, accelerating the thawing process is essential to minimize such damage. One effective approach is the use of materials capable of either rapid temperature elevation or suppression of ice recrystallization. In this work, we present a peptide-functionalized photothermal platform that integrates NIR-triggered accelerated thawing with localized ice recrystallization inhibition. The peptides directly exert IRI activity at the ice interface, complementing the photothermal heating effect. This dual-function strategy enables synergistic mitigation of damage, resulting in markedly improved post-thaw cell viability compared with photothermal heating alone.
