Biodegradable polyhydroxyalkanoates (PHAs) nanoparticles are fabricated via an emulsification and evaporation process. This involves forming emulsion droplets of PHA-dissolved chloroform in water through bulk mixing, followed by a high shear-driven droplet size reduction with a narrow size distribution. The diameter of the PHA nanoparticles is precisely controlled, ranging from 50 nm to 200 nm, by adjusting the pressure, the number of cycles, and the PHA concentration. The physical properties of the nanoparticles can be tailored based on the monomer ratio of 3-hydroxybutyrate to 4-hydroxybutyrate. Size-controlled PHA nanoparticles can encapsulate and deliver both lipophilic and hydrophilic active ingredients, such as ceramides and niacinamide, to the skin at the cellular level. To enhance biodegradability and reduce the cytotoxicity of the nanoparticles, we also introduce a PHA-based surfactant with poly(ethylene glycol) and polysaccharides instead of using conventional surfactants.
