Synthesis of an ionic aromatic cyclodextrin for a novel host molecule of adapalene

Adapalene, a third-generation topical retinoid, effectively treats acne vulgaris by modulating cell differentiation, keratinization, and exerting anti-inflammatory effects. However, its clinical use is limited by poor water solubility and potential skin irritation. Cyclodextrins (CD), cyclic oligosaccharides, can form inclusion complexes with hydrophobic drugs such as adapalene. Here, we report a novel synthetic CD to improve the host-guest interaction with adapalene via additional aromatic and electrostatic interactions. Adding a synthetic CD improved the solubility of adapalene in an aqueous solution, potentially reducing side effects and overcoming limitations.
