Stem Cell Spheroid Preparation and their Preservation Strategy using Biocompatible Polymers

In this study, we investigated the recovery of stem cell spheroids (SCSs) after cryopreservation at –196 °C for 7 days. The SCSs were pretreated with low-molecular-weight PEG of different molecular weights (200, 400, and 600 Da) for different time interval. We found that the recovery of the SCSs was significantly influenced by both the molecular weight of PEG and the preincubation time. When SCSs were preincubated their recovery rate was notably improved. It seems that the PEG diffuses into the SCSs, which not only prevents osmotic pressure buildup inside the cells but also inhibits ice formation. The recovered SCSs showed the ability to fuse and proliferate, and they could differentiate just as well as SCSs recovered after being frozen with a 10% dimethyl sulfoxide solution. This study suggests that simply preincubating the SCSs with this polymer significantly enhances their recovery after cryopreservation.
