3D Modeling of Mechanically Stimulated Abnormal Scars to Identify Therapeutic Targets

Abnormal scars are excessive forms of scarring that arise from an abnormal healing response, leading to conditions such as hypertrophic scars or keloids. Since full cure of the abnormal scars is challenging and often requires a combination of therapies for optimal management, understanding abnormal scar formation is crucial for developing effective treatments. In particular, mechanical stimuli, especially tension, are increasingly considered as an important factor facilitating the progression of abnormal scars. Here, we constructed a micrometer punching-based 3D matrix wound model with 3D collagen hydrogel to mimic the dermal layer, and applied compressive prestress to provide mechanical stimulation. Tension induces cellular stretching and cytoskeletal rearrangement, providing an opportunity to induce excessive cell activation and extended healing periods. This 3D model could be a versatile tool for identifying therapeutic targets for abnormal scars under mechanical tension.
