Advanced Glioblastoma-Targeted Aptamer Discovery by 3D Cell-SELEX with Hyaluronic Acid Hydrogel

Glioblastoma, a highly aggressive brain cancer, poses significant treatment challenges due to its invasive nature and therapy resistance. Aptamers can be a promising therapeutic agent for the glioblastoma due to their potential for highly specific targeting and enhanced diagnostic capabilities. They are identified by SELEX (Systematic Evolution of Ligands by Exponential enrichment), a directed evolution technique. However, traditional SELEX methods using 2D cultures do not accurately represent natural 3D tumor microenvironment with accumulated extracellular matrix (ECM). We employed hyaluronic acid (HA) hydrogel-based 3D cell culture model to better mimic in vivo glioblastoma microenvironments, and several aptamers were identified that specifically bind to the glioblastoma cell line. This 3D cell-SELEX may enhance the relevance and applicability of aptamer selection for glioblastoma, potentially leading to more effective diagnostic and therapeutic strategies.
