Drug Delivery System for Regenerating Permanently Damaged Skin Tissue

Skin wounds are common injuries and there is currently no method to regenerate permanently damaged skin. We introduce a microneedle drug delivery system using 1,4-dihydrophenonthrolin-4-one-3-carboxylic acid (1,4-DPCA) to regenerate damaged skin. The drug inhibits the enzyme prolyl hydroxylase, which suppresses the proliferation of HIF-1α cells, thus inducing artificial tissue regeneration. Microneedles have advantages like sustained transdermal drug delivery and minimal invasiveness, contributing to the maximization of regeneration effects. We synthesized microneedles by copolymerizing 1,4-DPCA with the biocompatible monomer 1-vinyl-2-pyrrolidone (VP) using UV-induced polymerization. For successful copolymerization, we attached an allylamine functional group to 1,4-DPCA using HBTU coupling reactions, facilitating its chemical bonding with VP. This drug delivery system will enable non-professionals to effectively treat damaged tissues, promoting wound healing and tissue regeneration.
