Enhanced Transdermal Delivery of Aconite Root with HA-PLGA Nanoparticles for Pain Relief

Traditional pain management, which depends on medications and dose adjustments, often fails to address neurotoxicity, leading to insufficient pain relief and undesirable side effects. Aconite root (AR), a medicinal herb, contains Aconine, which alleviates pain by blocking neural channels. However, AR contains aconitine, a toxic substance that is reduced through heating. HPLC confirms optimal Aconine and aconitine content after 48 hours of boiling AR, with minimal toxicity. This study develops HA-PLGA/AR NPs for controlled transdermal delivery, improving therapeutic outcomes. Characterization through TEM, DLS, drug release test and Franz diffusion cell confirms morphology and drug skin penetration of HA-PLGA/AR NPs, validating effectiveness in neuropathic pain management. Furthermore, a neurite outgrowth test on paclitaxel-induced PC12 cells demonstrates the analgesic effect of HA-PLGA/AR NPs, indicating a potential to alleviate various neuropathies.
