Cell-Inspired Hydrogel Microcapsules with a Concentric Oil Layer for Enhanced Antioxidant Retention

In this study, we present cell-inspired hydrogel microcapsules featuring an interstitial oil layer, created using triple emulsion droplets as templates, to enhance antioxidant retention. We utilize ionic gelation for the hydrogel shell to avoid exposing the encapsulated antioxidants to free radicals commonly produced during photopolymerization. The interstitial oil layer acts as a stimulus-responsive diffusion barrier, enabling efficient encapsulation and retention of antioxidants by providing a suitable pH microenvironment until osmotic pressure is applied for on-demand release. Additionally, incorporating a lipophilic reducing agent in the oil layer triggers a complementary reaction with the antioxidant, similar to the nonenzymatic antioxidant defense system in cells, and further enhances antioxidant retention. We also demonstrate that lowering the storage temperature fully recovers and even boosts antioxidant activity by reducing the oxidation rate while maintaining the complementary reaction with the lipophilic reducing agent
