Oral delivery offers many benefits but is limited by the harsh conditions of the digestive tract, which inhibit activity and complicate targeted intestinal release. Here, we introduce multicompartmental hydrogel microspheres with concentric oil layers to enhance oral drug delivery efficiency. These microspheres are produced from triple emulsion droplets and feature an intermediate oil layer that separates two prepolymer phases, allowing precise tuning of each compartment's composition. We demonstrate that the oil layer protects encapsulated material from the stomach's acidic environment. The hydrogel core provides high stability even in the stomach's high osmotic conditions. By fine-tuning the shell composition, we develop capsules that release selectively according to the gut's pH conditions. These microspheres show considerable potential for the inner beauty market by facilitating targeted delivery to the gut and promoting gut health, closely associated with skin health.
