Theranostic Polymeric Prodrug nanoparticles for Renal Ischemia/reperfusion Injury

Ischemia/reperfusion(I/R) injury is an inevitable pathological event occurring during the organ transplantation. Immediately after reperfusion, cells produce large amounts of ROS such as H2O2. While H2O2 promotes angiogenesis at physiological levels, its overexpression leads to exacerbated inflammation, and contributes to organ dysfunction. Therefore, the elimination of excessive H2O2 is crucial for mitigating I/R injury. In this study, we developed poly(arylboronate methacrylate) (pBMA) nanoparticles as a nanotheranostic agent for renal I/R injury and decorated their surface with TUDCA to enhance targeting of inflamed tissues. We demonstrated that T-pBMAs(TUDCA-coated pBMA nanoparticles) preferentially accumulate at sites of inflammation, enhance ultrasound contrast by generating CO2 bubbles, and effectively prevent I/R injury in a H2O2-triggered manner. Based on these, we anticipate that T-pBMAs have excellent translational potential.
