Polylactic Acid-based Multifunctional Porous Microspheres Loaded with Oncolytic Spores for Imaging-guided Cancer Therapy

Oncolytic bacteria are promising for targeted cancer therapy due to their selective germination in hypoxic tumor sites but delivering them effectively remains challenging. This study developed innovative multifunctional porous microspheres (MPMs) to address this issue. Biodegradable MPMs were coated with a positive polymer and loaded with negatively charged oncolytic spores, allowing targeted delivery to tumor sites using a magnetic field. This approach minimizes potential side effects. In vitro tests confirmed successful spore encapsulation and release, demonstrating their ability to activate and kill tumor cells while triggering an immune response against cancer. This novel strategy combining targeted delivery and spore activation holds great promise for advancing imaging-guided cancer therapy and improving patient outcomes.
