An Electro-Ferroptotic Nanocomplex for Spatiotemporally Controlled Cancer Ferroptosis via Irreversible Electroporation

Ferroptosis, an iron-dependent programmed cell death pathway, has emerged as a promising approach for cancer treatment. However, challenges such as low selectivity and uncontrollable release of intracellular Fe2+ have hindered its therapeutic application. This study presents a novel approach combining iron oxide nanoassembly (IONA), ascorbic acid-loaded liposomes (Lip-AA), and irreversible electroporation (IRE) to overcome these limitations. IONA and Lip-AA form an electrostatically stable electro-ferroptotic nanocomplex (EFN). Upon exposure to IRE, AA is released, reducing IONA to Fe2+, while IRE simultaneously increases tumor cell membrane permeability. This nanocomplex demonstrates synergistic effects, inhibiting tumor growth and increasing survival rates. This innovative approach represents a promising direction for next-generation tumor therapy, potentially improving treatment efficacy and patient outcomes.
